What kind of tools and techniques can be used for route tuning ?

There is JMX stats about the route performances.

FuseHQ can also shows performance and health stats etc. Its a management and monitoring tooling.

Likewise Fuse IDE can also show route performance stats. Look out for the upcoming Fuse IDE 2.1 release.
